The 17.2T preclinical MRI scanner, running under the responsibility of L. Ciobanu from the “NeuroPhysics” research group, is one of the highest magnetic field horizontal bore preclinical system in the world. The clear bore inside the gradients (85 mm) allows the imaging of small animals (rats, mice). Besides the very high magnetic field, another feature of this system is the high strength of the magnetic field gradients, capable of reaching 1000 mT/m. With these gradients, it is possible to acquire very high spatial resolution images (25 µm isotropic). The 17.2T system is equipped with a variety of radiofrequency coils: surface, phased array and volume coils for rat and mouse brain imaging, as well as with microcoils with diameters between 700 µm and 2 mm for imaging small samples. X nuclei surface coils (13C, 31P) for spectroscopy studies are also available.

The system will be soon equipped with the AVANCE NEO electronics that offers improved shimming performances, faster gradient switching times and gradient linearity corrections ensuring better image quality. Secondly, a cryo-platform and a cryo-cooled deuterium coil (2H) will be purchased to open the way to novel explorations of metabolic events occurring in the brain. A helium recovery and liquefaction system will be installed to eliminate the need for helium supplies.

While <sup>1</sup>H<sub>2</sub>O is the focus of almost all MRI studies, several other molecules and ions exist whose NMR signals are much more informative regarding the biophysical, metabolic or physiological status of those tissues. Over the past years, I used <sup>1</sup>H and <sup>31</sup>P NMR Spectroscopy to quantify metabolites involved in neurotransmission and metabolism, such as neurotransmitters Glutamate and GABA or energy store/substrates Adenosine Tri-Phosphate and Lactate. Using dedicated fast ultra-short echo-time imaging sequences, I also investigated Sodium (<sup>23</sup>Na) ions whose transmembrane concentration gradient is maintained at great energy cost and plays a major role in establishing the neurons’ resting potential.

One of my recent research line is Lithium-7 MRI. Lithium salts are the main treatment for bipolar disorder, yet the mechanism of action remains a mystery, and 7Li MRI is the only technique allowing lithium non- invasive mapping and quantification in the brain. Dynamic <sup>13</sup>C or <sup>2</sup>H NMR are very interesting non-irradiating alternatives to \[18F]FDG-PET benefitting from their greater chemical/spectral specificities notably to discriminate and quantify aerobic vs. anaerobic energy synthesis pathways, neuronal and glial metabolism or even probe neurotransmission via the GABA and Glutamate synthesis and recycling rates. All of those weak signals benefit from the extremely high magnetic fields available at NeuroSpin enabling their investigation at better temporal and spatial resolutions.

My goal is to advance those methods and collaborate with biologists, physicians, bio-informaticians and neuroscientists to incorporate them within multimodal clinical and preclinical research protocols to better understand psychiatric and neurological diseases, their eventual treatments, and to develop novel biomarkers of disease progression or response to treatment.

In January 2024, Alfredo Lopez-Kolkovsky joined the CICLOPS group to accelerate the development of metabolic neuroimaging in particular on the 11.7T Iseult scanner.

To be able to better model drug delivery, we have measured the diffusion properties of nanoparticles after their crossing of the BBB. On the technological side, we have implemented passive cavitation detection and feedback loop monitoring at high frame rate to ensure safety of the ultrasound procedure with a patented approach. We also introduced safe large volume BBB opening sequences for applications such as whole brain or hemispherical drug delivery.

Regarding applications, we have demonstrated the interest of the technique in small animal models, mainly in neuro-oncology, Alzheimer’s disease and gene delivery fields. We showed the capabilities to increase significantly the local concentration of MRI contrast agents and PET tracers, of drug-loaded nanoparticles, of therapeutic antibodies and of adeno- associated viruses. To translate this to patients, we have built a public-private French consortium to design, assemble, and test a new prototype of clinical device. This system is robotized and neuro-navigated to make
it widely spreadable in hospitals. It is cost-effective and compatible with short ambulatory procedures in order to perform chronic sessions over months or years. During the course of this project, every technological feature has been validated in non-human primates. We now want to pursue its clinical development through a spin-off company.
